Fabrication and Properties of Poly(vinylalcohol)-glycosaminoglycantype I Collagen Composite Membrane as Tissue Regeneration Scaffolds

The objective of this paper is to design a porous polyvinyl alcohol (PVA) based on composite membrane with certain mechanical strength and biocompatibilities serving as tissue regenerative scaffolds. PVA-glycosaminoglycan (GAG)-type I collagen(COL) composite membrane was fabricated by PVA with different molecular weight (Mw) and alcoholysis degree (AD) being blended with certain amounts of GAG and COL and dried at 38℃ for 24 h. The water content of the composite membranes were from 61.9% to 95.1% and swelling ratio ranged from 123.6% to 621.7%. Scanning electron microscope (SEM) analysis proved that PVA-GAG-COL composite membrane has porous and homogenous structure. Biocompatibility test results showed that the composite membrane was nontoxic, which could promote adhesion and proliferation of fibroblasts on the composite membrane. In conclusion, PVA-GAG-COL composite membrane with high water content and swelling ratio, suitable mechanical strength and good biocompatibility, has potential in tissue engineering and regenerative medicine.

Analysis of TNF-α-induced Leukocyte Adhesion to Vascular Endothelial Cells Regulated by Fluid Shear Stress Using Microfluidic Chip-based Technology

This paper aims to the research of the impact of fluid shear stress on the adhesion between vascular endothelial cells and leukocyte induced by tumor necrosis factor-α(TNF-α) by microfliudic chip technology. Microfluidic chip was fabricated by soft lithograph; Endothelial microfluidic chip was constructed by optimizing types of the extracellular matrix proteins modified in the microchannel and cell incubation time; human umbilical vein endothelial cells EA.Hy926 lined in the microchannel were exposed to fluid shear stress of 1.68 dynes/cm
2
and 8.4 dynes/cm
2
respectively. Meanwhile, adhesion between EA.Hy926 cells and leukocyte was induced by TNF-αunder a flow condition. EA. Hy926 cell cultured in the static condition was used as control group. The numbers of fluorescently-labeled leukocyte in microchannel were counted to quantize the adhesion level between EA. Hy926 cells and leukocyte; cell immunofluorescence technique was used to detect the intercellular adhesion molecule (ICAM-1) expression. The constructed endothelial microfluidic chip can afford to the fluid shear stress and respond to exogenous stimulus of TNF-α; compared with the adhesion numbers of leukocyte in control group, adhesion between EA. Hy926 cells exposed to low fluid shear stress and leukocyte was reduced under the stimulus of TNF-α at a concentration of 10 ng/ml(

and provide a new technical platform for exploring the pathophysiology of the related cardiovascular system diseases under a flow environment.

The Effect of Laser Acupuncture on the Levels of IL-2, IL-6 and TGF-β1 in Serum of Rats with Atrophic Gastritis

Objective:
To study the effect of three different methods such as medicine, acupuncture and laser acupuncture on the levels of interleukin 2 (IL-2), interleukin 6 (IL-6) and transforming growth factor-β1 (TGF-β1) in rat's serum with chronic atrophic gastritis.
Methods:
9 rats were randomly selected from 60 rats as normal control group, and the other rats were used to replicate the animal model of chronic atrophic gastritis with comprehensive method, 5 rats were randomly inspected at the 8th week in the process of creating model for gastric mucosa pathological examination; when gastric mucosa shows CAG symptoms such as various degree of congestion, bleeding, atrophy, metaplasia, the model rats were randomly divided into model group, medicine group, acupuncture group and laser acupuncture group. The rats of medicine group were chronically administered with yanshenjianwei capsule and Kangfuxin Liquid; the groups of acupuncture and laser acupuncture selected Zusanli acupuncture as acupuncture points,after the treatment for 14 d, blood was removed, double antibody sandwich enzyme linked immunosorbent assay (ELISA) was used to detect the levels of IL-2 , IL-6 and TGF-β1 in serum.
Results:
Compared with normal group, the contents of IL-2, IL-6 and TGF-β1 in serum of model rats increased (
P
<0.05 or

<0.01). Medicine, acupuncture and laser acupuncture could reduce the contents of IL-2, IL-6 and TGF-β1, IL-2 in serum of the medicine group and laser acupuncture group was significantly decreased (

<0.05); IL-6 in acupuncture group and laser acupuncture group were significantly decreased (
P
<0.05 or
P
<0.01);and the most significant is laser acupuncture group (
P
<0.01). Medicine, acupuncture and laser acupuncture could significantly reduce TGF-β1 content in serum (

P
<0.01), and the particularly significant was laser acupuncture(

<0.01).
Conclusion:
Laser acupuncture has the function of reducing the inflammatory response and adjusting immune function. It can effectively reduce the expression level of IL-2, IL-6 and TGF- β1 in serum of rats with chronic atrophic gastritis, and its effects are better than traditional acupuncture and medicine.

Single Channel Fetal ECG Detection Using LMS and RLS Adaptive Filters

ECG is an important tool for the primary diagnosis of heart diseases, which shows the electrophysiology of the heart. In our method, a single maternal abdominal ECG signal is taken as an input signal and the maternal P-QRS-T complexes of original signal is averaged and repeated and taken as a reference signal. LMS and RLS adaptive filters algorithms are applied. The results showed that the fetal ECGs have been successfully detected. The accuracy of Daisy database was up to 84% of LMS and 88% of RLS while PhysioNet was up to 98% and 96% for LMS and RLS respectively.

The Influence of RNA Secondary Structure on the Efficiency of siRNA Silencing

In the application of RNAi technology, it is an essential step to design siRNA applicable to target gene. At present, there are many researches and conclusions on siRNA design. This paper aims to the influences of mRNA secondary structure or siRNA antisense-strand secondary structure on siRNA silence efficiency. The paper also discusses the problems and sets out further insights in the research.
