93 Antifungal Mechanism of the Combination of Eugenol with Ketoconazole Against Malassezia

Malasseria specics are common pathogenic fungus that causes several skin diseases, which have a certain rate of resistance to antifungal drugs. Attention has been brought to aromatic compounds because of their promising antimicrobial properties. The present study aims to investigate the mechanism of the anti-Malassezia activity of eugenol and to evaluate its' potential synergism with ketoconazole. The minimum inhibitory concentration (MIC) of eugenol and ketoconazole were investigated using a standard microbroth dilution assay. It was found that eugenol and ketoconazole have a synergistic effect with the FICI value 0.5 through the checkerboard assay. The effects of eugenol and its' combination with ketoconazole on the cell structure (SEM, TEM), the cell wall (sorbitol protection effect), and the cell membrane (the release of intracellular material, complex with the ergosterol, ergosterol synthesis) were investigated. The results of these studies on the mechanisms of action suggested that eugenol exerts antifungal effects on the cell wall and cell membrane of M. furfur, the biosynthesis of ergosterol was also being disturbed. The destruction was strengthened when combined with ketoconazole.

105 Determination of Anisotropic Factors of Pyrolytic Carbon by Electron Diffraction

The preferred orientation of pyrolytic carbon coating is an important performance parameter for the safe use of artificial mechanical heart valve.In this paper, the selected area electron diffraction (SAED) analysis of pyrolytic carbon coating samples of artificial heart valve was carried out by transmission electron microscopy (TEM), and the experimental method of characterizing the preferred orientation of pyrolytic carbon by electron diffraction spectrum, namely bacon anisotropy factor (BAF), was described, and the orientation angle and BAF corresponding to the electron diffraction spectrum were measured.The results show that the BAF value can directly reflect the anisotropy of pyrolytic carbon in the selected area. The BAF value range is from 1 to infinity, and the higher the preferred orientation is, the greater the value is. The BAF value decreases exponentially with the increase of orientation angle. When the pyrolytic carbon tends to be isotropic, the orientation angle tends to 180 and the BAF value tends to 1.

111 The Access Management of Medical Consumables in Public Hospitals

Objective: To strengthen the standardized management of hospital medical consumables, combined with national policies and regulations, to explore the access process of medical consumables in line with hospital process management. Methods: By formulating the management system of medical consumables, establishing medical consumables applied through different channels, adopting appropriate audit standards and optimizing different application processes, we can better refine the management of consumables access. Results: The establishment of the standard can well cover the access audit of all medical consumables to be admitted to the hospital. Conclusion: By strengthening the audit of medical consumables before entering the hospital, the access principle of medical consumables is well standardized, the repeated admission of medical consum ables is reduced, and the unnecessary admission of medical consumables is avoided.

118 Visualization Analysis of CRISPR Gene-editing Knowledge Map Based on CiteSpace

CRISPR is an adaptive immune defense system found in bacteria and archaea that is resistant to heterologous invasive genetic material. Later studies showed that the CRISPR system can be used for gene editing. This study used the Web of Science database as a search object, then visually analyzed the literature related to CRISPR gene-editing technology with CiteSpaceV. The results show that publications had increased year by year. USA ranked first in terms of publications, China is second, but the centrality is very low. Doudna JA and Zhang F have made outstanding contributions. There are close connections between the intermnal institutions of the various states, but there are few links between the states. The hot spot and frontier are the application of CRISPR in animals, plants, detection, diagnosis, and clinical treatment.
